Patolojik Prion Proteininin Tespiti

Abstract

Bu makalede, prion hastalıklarının tanısında kullanılan güncel yöntemler hakkında bilgiler verilmektedir. Prion hastalıkları, patolojik prionların neden olduğu nörodejeneratif hastalıklardır. Patolojik prionlar (PrPSc), konak tarafından kodlanan prion proteininin (PrPC), anormal izoformlarıdır. PrPC'nin, PrPSc'e spontan dönüşümü, nöronlarda ve farklı dokularda (beyin dokusu, lenf bezleri ve tonsiller gibi) PrPSc birikimine yol açmaktadır. Prion hastalıklarının tanı yöntemlerinin birçoğu, nöronlarda ve dokulardaki PrPSc'nin tespitine dayanmaktadır. Güvenilir bir tanı metodunun ön koşulu, birçok örnekte PrPSc'i yüksek oranda tespit etmesidir. Bu amaç doğrultusunda, PrPSc'i tespit etmek için çeşitli yöntemler ve prion enfeksiyonuna duyarlı hücre hatları geliştirilmiştir. This article gives information about the current methods used in diagnosis of prion diseases. Prion diseases are neurodegenerative disorders caused by pathogenic prions. Pathogenic prions are abnormal isoform of prion protein (PrPC), is a host-encoded molecule. PrPC to PrPSc spontaneous conversion leads to PrPSc accumulation in neurons and different tissues (such as brain tissue, lymph nodes and tonsils). Most of the diagnostic methods for prion diseases are dependent on PrPSc detection in neurons and tissues. High rate PrPSc detection in many different samples is a prerequisite for a reliable diagnostic method. For this purpose, several methods and cell lines that are susceptible to prion infection have been developed for detection of PrPSc.
